Apimostinel is a drug that has been studied for its effects on depression. It is an allosteric modulator of the mTOR complex 1, which is a protein involved in cell growth and death. Apimostinel binds to the rapamycin-binding domain of FKBP12 and inhibits the interaction with rapamycin, thereby inhibiting the mTOR complex 1. This inhibition leads to increased release of glutamate from nerve cells, which may lead to antidepressant effects. Apimostinel also has synergistic effects with tuberculosis drugs such as isoniazid and rifampin, which have been shown to be effective against multidrug-resistant tuberculosis (MDRTB).
