Afuresertib hydrochloride is a potent and selective inhibitor of histone deacetylases (HDACs). It has been shown to inhibit the growth of prostate cancer cells in vitro and in vivo, which is due to its ability to decrease the expression of genes related to tumour growth. Afuresertib hydrochloride has also been shown to be effective as an oral treatment for prostate cancer resistant to castration therapy. This drug inhibits HDAC activity by binding irreversibly to the enzyme, preventing it from deacetylating histones and other proteins. The mechanism of action for this drug is not fully understood but may be due to its ability to activate the transcriptional co-repressor p300 and increase the expression of genes that are involved in cell cycle control.
