AP5 is a peptide that belongs to the class of activator peptides. It is a potent inhibitor of potassium channels, and has been shown to inhibit calcium channels as well. AP5 is an ion channel blocker that blocks the opening of potassium channels in response to membrane depolarization, preventing the flow of ions through the cell membrane. The binding affinity for AP5 is high and it can be used as a research tool or pharmacology agent.
AP5 binds to the extracellular domain at a site known as S6 on the ligand-binding domain of voltage-gated potassium channels, which blocks ion flow through these channels. This prevents depolarization of the neuron and therefore prevents transmission of nerve impulses.
AP5 binds with high affinity to both T-type calcium channels (IC50 = 0.4 nM) and N-type calcium channels (IC50 = 1 nM).
