Ceramides are a family of sphingolipids that play an important role in cell membrane function. They are synthesized from fatty acids and glycerol-3-phosphate by the enzyme dihydroceramide synthase. Ceramides are flexible molecules that can change shape, allowing them to form complexes with other lipid molecules. This flexibility also allows them to bind to proteins at different sites on the protein surface, as well as inside the protein's structure. The c2 ceramide is a synthetic analogue of the natural ceramide with two double bonds between carbons 17 and 18. It is an inhibitor of ion channels, such as nicotinic acetylcholine receptor (nAChR), alpha-7 nicotinic acetylcholine receptor (α7 nAChR), and voltage-gated potassium channels (Kv1.2).
