The small molecule RGD peptidomimetic avb3 integrin antagonist XJ-735 contains both anionic and cationic binding sites that straddle the RGD binding domain of the avb3 integrin. The IC50 values of XJ 735 for avb3-mediated inhibition of human and porcine cell adhesion, and vascular smooth muscle cell migration, ranged from 0.6 to 4.4 uM. In contrast, IC50 for porcine or human aIIb/b3, a4b1, avb5, and a5b1 inhibition exceeded 100 uM.
