Mevalonic acid lithium salt is a drug that inhibits the activity of the enzyme mevalonate kinase and is used to treat bowel disease. Mevalonic acid lithium salt has been shown to inhibit the production of inflammatory cytokines such as TNF-α, IL-1β, and IL-6 in response to Toll-like receptor (TLR) activation and has potential therapeutic effects on Crohn's disease. This drug also inhibits protein synthesis in a number of different cell types including human neutrophils and macrophages, which may be due to its ability to suppress TLR signalling pathways. Mevalonic acid lithium salt binds to the cell nucleus and alters the expression of nuclear proteins such as histones. This binding also prevents transcriptional activation by blocking RNA polymerase from accessing DNA.
