Norfenfluramine hydrochloride is a drug that has been used to treat obesity. Norfenfluramine inhibits the reuptake of serotonin, thereby increasing its concentration in the brain. The clinical efficacy of norfenfluramine in treating obesity was due to this action on serotonin. Norfenfluramine has also been shown to cause primary pulmonary hypertension and cardiac valvular disease, which may be due to its ability to activate 5-HT2 receptors and induce mitochondrial membrane depolarization. This drug also interacts with non-steroidal anti-inflammatory agents by competition for binding at the 2-aminoethoxydiphenyl borate site of cyclooxygenase-1 (COX-1). 5-HT2C agonism may play an important role in the development of these side effects.
