Nω-Acetylhistamine is a histamine antagonist that blocks the effects of histamine on the central nervous system and peripheral vessels. It also has been shown to inhibit microbial metabolism in x-ray crystal structures and model systems, with an IC50 value of 0.5 mM. Nϋ-Acetylhistamine inhibits the activity of the enzyme h1 receptor, which leads to decreased energy metabolism and increased low density lipoprotein cholesterol levels. This compound is not metabolized by mammalian cells due to its lack of amide group, which makes it a more potent inhibitor than other h1 antagonists such as pyrilamine (IC50 = 1 mM). The mechanism of action for this drug is based on intermolecular hydrogen bonding, which stabilizes its binding site in the active site of h1 receptors.
