5-Hydroxyisoindoline-1,3-dione is a novel HDAC inhibitor that has been shown to inhibit the activity of human prostate cancer cells. This drug binds to the catalytic site of the HDAC enzyme and prevents acetylation of histones, leading to an increase in transcriptional activity for genes involved in cell proliferation and differentiation. 5-Hydroxyisoindoline-1,3-dione also has a carbonyl group that may be responsible for its selectivity against HDAC2 enzyme. The carbonyl group is thought to interact with the substrate binding site of HDAC2, which would account for its selectivity over other HDAC enzymes. The 5-hydroxyisoindoline moiety is substituted with a hydroxamic acid group that can potentially react with DNA and form a covalent bond. This interaction leads to inhibition of transcriptional activity by preventing acetylation of histones.
