4-Methoxy-5-(3-morpholinopropoxy)-2-nitrobenzonitrile is a synthetic intermediate for the synthesis of gefitinib, an anticancer medicine that is used to treat non-small cell lung cancer. The compound reacts with chloro-, 3-chloro-4-fluoroaniline and benzamide in the presence of phosphoric acid to produce the desired product. The reaction proceeds through a transfer hydrogenation mechanism, which is catalyzed by palladium chloride. This process produces 4-methoxy-5-(3-morpholinopropoxy)-2-nitrobenzonitrile at high yield and with excellent purity. It has been shown that the reaction intermediates can be salified with hydrochloric acid or hydrobromic acid to form a stable salt.
