Gefitinib is a receptor tyrosine kinase inhibitor that targets the epidermal growth factor (EGF) receptor and the HER2/neu receptor. Gefitinib binds to the ATP-binding site of tyrosine kinases and inhibits their activity, which prevents the activation of downstream signalling pathways. This drug has been shown to inhibit cancer cell proliferation in vitro and to inhibit tumor growth in vivo. Gefitinib also acts as an antioxidant by inhibiting lipid peroxidation, thereby blocking one of the mechanisms by which cancer cells proliferate. The drug is orally bioavailable, with a half-life of 6 hours.
Gefitinib has been shown to inhibit the transduction of epidermal growth factor (EGF) from its membrane receptor on the surface of cells to its cytoplasmic domain. It also blocks EGF-induced activation of protein kinase B, which is necessary for mitogenesis and cell survival
