5-Carboxy-fluorescein diacetate N-succinimidyl ester (CFSE) is a fluorescent probe that is used in cell biology to measure proliferation and differentiation. CFSE binds with high affinity to basic proteins, such as albumin or transferrin, and can be used to detect the presence of these proteins in cells. CFSE can also be used as a fluorescent probe for polymerase chain reaction (PCR) analysis because it binds to the DNA polymerase during PCR amplification. The fluorescence intensity of CFSE is proportional to the number of molecules bound. This dye has been shown to inhibit leukemia inhibitory factor (LIF), which may be due to its ability to act as an immunosuppressant.
