N3-PEG5-CH2CH2COOH is a wst-1 prodrug that has been designed to penetrate the blood-brain barrier. It has an anionic head group and a hydrophilic tail, which allows it to be taken up by cells in the tumor. N3-PEG5-CH2CH2COOH has been shown to induce cell death in glioma cells through the inhibition of protein synthesis. N3-PEG5-CH2CH2COOH also inhibits cancer cell proliferation, survival and invasion, and induces apoptosis. The prodrug is converted into its active form, propargylamine, which binds with chloride ions to produce reactive oxygen species (ROS) that cause DNA damage and ultimately death of cancer cells.
The prodrug can also be pegylated for increased stability and efficacy.
