1-Methyl-2-iodo-4-hydroxybenzene is a potent inhibitor of fungal polymerases. It binds to the active site of the enzyme, which contains a cysteine residue, and inhibits mammalian DNA polymerase activity. The inhibition of mammalian DNA polymerase by 1-methyl-2-iodo-4-hydroxybenzene occurs at concentrations below those needed for inhibition of fungal DNA polymerases. The potency is significantly lower than that of talaromycin or dehydroaltenusin, which are inhibitors of polyketide synthases. This drug also has inhibitory effects on Alternaria and Talaromyces fungi, and may be used as an antifungal agent.
