T863 is a cell-permeant, small molecule inhibitor of fatty acid synthase. It has been shown to reduce the size of fatty liver in mice and improve glucose tolerance in diabetic animals. T863 inhibits fatty acid synthesis by binding to the carboxyl group on the enzyme's active site and blocking the esterification of malonyl-CoA to acetyl-CoA. T863 also decreases diacylglycerol synthesis and increases protein synthesis in liver cells. This agent has pharmacokinetic properties that allow it to be administered orally, which may make it an attractive treatment option for patients with hepatic steatosis or type 2 diabetes mellitus.
