MI 503 is a retinoid that belongs to the group of all-trans retinoic acid. It has been shown to inhibit leukemia cell proliferation in vitro and in vivo. MI 503 induces apoptosis by binding to the epidermal growth factor receptor (EGFR), which leads to the activation of downstream signaling pathways. These pathways include phosphatidylinositol 3-kinase, protein kinase B, and extracellular signal-regulated kinases 1 and 2, which eventually lead to activation of caspases and cleavage of poly-(ADP-ribose) polymerase.

MI 503 also inhibits the activity of Toll-like receptors (TLR), which are proteins that recognize pathogen-associated molecular patterns on cells or molecules outside of cells. TLRs play an important role in regulating inflammation by controlling cytokine production, as well as promoting tumorigenesis.
