CA-4948 is a new bicyclic heterocycle that has been shown to have a high affinity for the toll-like receptor TLR4. CA-4948 has been found to be well tolerated in vivo and in vitro, with no tumorigenicity observed in mice. CA-4948 induces an antitumor response through the activation of hematopoietic cells and macrophages. This drug also activates TLR4 in the submandibular gland and microenvironment, leading to an increased production of proinflammatory cytokines such as IL-6, TNFα, and IL-1β. These cytokines promote an antitumor response by inducing the recruitment of cytotoxic CD8+ T cells.
