2-Amino-4-phenylthiazole hydrobromide monohydrate (2APT) is an inhibitor of histone deacetylase. It was synthesized by reacting 2-amino-4-phenylthiazole with hydrobromic acid, and the resulting salt was obtained as a white solid. The structure of 2APT has been determined by X-ray crystallography. The compound is a hydrogen bond donor and a strong hydrophobic ligand, which may be responsible for its binding to the active site of HATs.
2APT inhibits the activity of HATs by forming a covalent bond with the catalytic cysteine residue in the active site. This inhibition leads to decreased acetylation of histones, which results in changes in gene expression and ultimately cell growth arrest or apoptosis.
