N-[3-(Benzyloxy)pyridin-2-yl]benzenesulfonamide (NBP) is a drug that has been shown to have analgesic properties in animal pain models. NBP has been shown to bind to a proton receptor, which may be localized in the blood-brain barrier. It is speculated that this binding facilitates the passage of protons from the blood into the brain, leading to analgesia. In addition, NBP has been shown to have an affinity for interstitial fluid and facilitation of cerebrospinal fluid. This facilitation may lead to increased water permeability across the blood-brain barrier by increasing astrocyte expression and allowing for more water channels. The hypothesized mechanism of action is that NBP binds to a facilitator receptor on astrocytes, which leads to increased expression of aquaporin 4 (AQP4), which increases water permeability across the blood-brain barrier.!--
