Ondansetron is a drug that is used to prevent nausea and vomiting. It is an antagonist at the 5-HT3 receptor and blocks the actions of serotonin on this receptor. Ondansetron has a high antitumor activity and can be used for the treatment of tumors in humans, such as cancers of the breast, lung, stomach, colon, and pancreas. This drug also promotes cell death by inhibiting DNA synthesis or preventing cell division. Ondansetron binds to polymerase enzyme by forming hydrogen bonds with its active site. The pyridine ring of ondansetron interacts with the polymerase's nucleotide binding site and prevents it from releasing a nucleotide (a component of DNA) back into solution. This action prevents DNA synthesis and thus inhibits cancer growth as well as tumor cell division.
