Telaprevir is a synthetic amide that has been shown to be an effective drug against hepatitis C. It is a prodrug that is converted in vivo to its active form, telaprevir diphosphate (TP). Telaprevir inhibits the RNA-dependent DNA polymerase, which prevents the virus from replicating. It can also inhibit other enzymes involved in HCV replication, such as NS5A protein and NS3 protease. The synthesis of telaprevir begins with condensation of cyclopropylamine and 2-hydroxy-N-cyclopropylhexanamide with pyridine to form an imine intermediate. The imine is then hydrogenated by catalytic reduction to form the desired product. Telaprevir was developed using a novel synthetic method that does not require protection or deprotection steps.
