Complement factor B is a vital factor in the alternative pathway of the complement system and is involved in the formation of the C3 convertase, a crucial activator of the complement cascade. The alternative pathway is activated by auto-activation in which complement 3 (C3) becomes hydro-lysed. When binding to hydrolysed C3 factor B is cleaved by factor D. From the products Ba and Bb, generated from this cleavage, Bb makes up the C3 convertase (C3bBb). Due to its serine protease domain Bb has the ability when associated with hydrolysed C3, to cleave C3 molecules to produce the C3b in the C3 convertase. Factor B can be used as a marker of the alternative pathway in re-search.Overall the main functions of the complement system is to mark cells for phagocytosis, the recruitment of inflammatory cells and the cell lysis of bacteria cell by the MAC. Anaphylatoxins produced recruit neutrophils and causes the inflammatory response while the MAC produces pores in the bacterial membrane thus causing a Ca2+ influx into the cell and bacterial cell death.The inappropriate activation of the alternative pathways could contribute to diseases such as spon-taneous fetal loss in mice, macular degeneration and Asthma. The complement system as a whole can be associated with- neurological diseases, bacterial meningitis, thrombotic disorders, and autoimmune diseases.The proline residue at position 10 is isotopically labelled with carbon-13 (5) and nitrogen-15 (1).
