SQ 109 is a deuterated analog of pyrazinoic acid, an inhibitor of bacterial cell wall synthesis. SQ 109 has been shown to have antimycobacterial activity against Mycobacterium tuberculosis and Mycobacterium bovis in vitro assays. It also disrupts the mitochondrial membrane potential and inhibits nuclear DNA replication. SQ 109 is a potent inhibitor of TB-specific target enzymes, such as pyrazinamidase, which blocks the conversion of pyrazinoic acid to pyrazinoate by blocking the enzyme that catalyzes this reaction. The drug has not been tested in vivo. SQ 109 has shown resistance to rifampicin and other antibiotics in resistant mutants.
