4-Chlorobenzyl isothiocyanate (4CIT) is an isothiocyanate that has been shown to have anticancer activity in cell culture and animal models. It inhibits the proliferation of cancer cells by causing DNA damage, inhibiting protein synthesis, and inducing apoptosis. This compound causes DNA damage by forming covalent bonds with nucleophilic groups of the cancer cells. The mechanism of action for 4CIT in cancer cells may be due to a chlorination reaction that produces reactive intermediates that react with nucleophilic groups on the surface of the cancer cell, leading to DNA damage. 4CIT also inhibits protein synthesis by reacting with sulfhydryl groups on enzyme cofactors. This leads to changes in cellular metabolism and ultimately causes cell death.
