Piperaquine is a 4-aminoquinoline compound that is used to treat malaria. Piperaquine tetraphosphate tetrahydrate (PQTP) is the active form of piperaquine and has been shown to have strong antimicrobial activity against both Gram-positive and Gram-negative bacteria. PQTP inhibits protein synthesis in bacterial cells by binding to the 30S ribosomal subunit, preventing peptide bond formation. The insoluble polymer coating of the microcapsules protects the drug from degradation by digestive enzymes, increasing its absorption and efficacy. PQTP has a pharmacokinetic half-life of approximately 20 hours in humans and can be administered orally or intravenously for treatment of severe malaria. The oral bioavailability of PQTP is around 80%.
