2-Chloro-4,6-diphenyl-nicotinonitrile (2C6DN) is a synthetic drug that has been shown to have antimalarial activity in vivo. It is highly effective against the plasmodium falciparum strain and has been shown to be a suppressive agent during maturation. 2C6DN binds to the quinoline site of the plasmodium enzyme dihydroorotate dehydrogenase, blocking orotic acid synthesis. This product also inhibits the development of chloroquine resistance in strains of plasmodium falciparum. 2C6DN does not inhibit mammalian DNA polymerases or RNA polymerases and is an analog of chloroquine.
