5-Nitro-2-phenyl-1H-indole is a nitrosopyrrole molecule that has been shown to inhibit the activity of efflux pumps, which are proteins that transport molecules out of cells. It is also an inhibitor of water uptake into cells and blocks uptake of drugs into multidrug resistant bacteria. This molecule has antibacterial properties and can be used in the treatment of tuberculosis. 5-Nitro-2-phenyl-1H-indole is synthesized by reacting butanedione with nitric acid, followed by nitration with concentrated sulfuric acid. This compound binds to the pump active site and inhibits its function by interacting with a water molecule that is crucial for the pump's operation. The binding prevents release of ions from the cell, inhibiting uptake and retention in bacteria.
