D-p-Hydroxyphenylglycinamide (d-pHPG) is a beta-lactam antibiotic that belongs to the class of monosubstituted, beta-lactam, amide antibiotics. It is a prodrug that is hydrolyzed in vivo to its active form. d-pHPG has been shown to inhibit the growth of bacteria such as Staphylococcus aureus and Escherichia coli. This drug binds to bacterial ribosomes and inhibits protein synthesis by binding to the 50S ribosomal subunit. The beta-lactam ring of d-pHPG interacts with the peptidyltransferase site on the ribosome, preventing peptide bond formation. The alkoxy groups on d-pHPG interact with the 30S ribosomal subunit and inhibit protein synthesis by inhibiting translocation of tRNA from 30S to 50S subunits.
