NU7026 is a protein kinase inhibitor that selectively inhibits the activity of DNA-PKcs, a protein kinase involved in the apoptosis pathway. NU7026 has been shown to induce apoptosis in cancer cells by inhibiting DNA repair and inducing cell cycle arrest. This drug also induces pluripotent stem cells to differentiate into neural progenitor cells, which may be due to its inhibition of DNA-PKcs and its downstream effects on the damage response. NU7026 is an ATP competitive inhibitor that binds to the ATP site of PKC and prevents ADP from binding, thereby preventing phosphorylation of proteins. The drug inhibits PKC at nanomolar concentrations, but has no effect on other protein kinases such as ERK1/2 or PKA.
