4-Amino-5-carboxy-2-ethylmercaptopyrimidine (ACEM) is a potent inhibitor of tumor cell proliferation. It has been shown to inhibit the growth of colorectal cancer cells in vivo and in vitro. ACEM inhibits the proliferation of human colorectal tumor cells, hCT116, which are resistant to 5FU and oxaliplatin. This drug also has inhibitory activities against cancer cells from other types of cancers, such as breast cancer and melanoma. The mechanism for this inhibition is not known, but it may be due to ACEM's ability to bind to RNA polymerase II and prevent transcription or its ability to block DNA synthesis by binding to DNA.
