4-Adamantylbenzenesulfonyl chloride is a conformationally constrained analogue of the natural product 1,2-dihydrobenzofuran. It is a potent inhibitor of human cytochrome P450 enzymes, with Ki values in the range of 2 to 10 nM. 4-Adamantylbenzenesulfonyl chloride binds to the active site of cytochrome P450 and blocks access to substrate, thereby inhibiting the metabolism of drugs such as phenytoin and propranolol. 4-Adamantylbenzenesulfonyl chloride can be used as a probe for investigating enzyme activity in vitro or in vivo.
