4-(5-Amino-1,3,4-thiadiazol-2-yl)benzoic acid (AZD0530) is a potent inhibitor of the mitochondrial matrix enzyme aconitase. Acetylcarnitine is an intermediate in this pathway and is involved in energy production by mitochondria. AZD0530 inhibits the activity of acetylcarnitine and therefore limits the amount of energy produced by mitochondria. This leads to a reduction in ATP and an increase in lactate levels. It has been shown that AZD0530 can reduce diabetic retinopathy by inhibiting the growth of vascular endothelial cells and improving blood flow to the retina. AZD0530 also reduces angiogenesis, which is the formation of new blood vessels from pre-existing ones, due to its ability to inhibit vascular endothelial cell proliferation.
