2-(7-Amino-2-oxo-2H-chromen-4-yl)acetic acid (CAA) is a fluorescent probe that can be used to identify the 14α-demethylase, which is an important enzyme in the synthesis of coumarin derivatives. CAA was found to be able to bind to the surface of lipid monolayers and form a nonfluorescent complex with this enzyme. This binding was specific, as no other esterase tested had any activity on CAA. Fluorescence microscopy studies have shown that CAA binds preferentially to the catalytic site of 14α-demethylase and can be used as a model system for studying this enzyme.
