- 3’-beta-Amino-3’-deoxy thymidine
It is a noncompetitive inhibitor of human DNA polymerase. It binds to the active site of the enzyme and inhibits enzymatic activity by binding to the substrate-binding site. The drug has been shown to inhibit transcriptase activity in herpes simplex virus and cytomegalovirus, as well as viral protein synthesis in HIV-1 infected cells. Although it is not active against RNA viruses, it does prevent the formation of stable complexes with dinucleotide phosphate that are necessary for viral replication. This drug has clinical relevance for the treatment of human immunodeficiency virus (HIV) infection and other diseases caused by DNA viruses, such as hepatitis B virus.
The drug inhibits human liver cells from transcribing dna, which prevents protein synthesis. It also binds to NADPH cytochrome P450 reductase, preventing it from reducing NADP+ into NADPH.
