(3-Amino-4-methyl-phenyl)-carbamic acid tert-butyl ester is a tyrosine kinase inhibitor that binds to the ATP binding site of b-raf. This drug has been shown to be selective against v600e mutation, which is found in chronic myeloid leukemia (CML) cells that are resistant to other tyrosine kinase inhibitors. The selectivity profile of this drug is mediated by its allosteric binding profile and its conformation. It inhibits the kinases that phosphorylate the residue Y1130 in c-Raf, leading to
decreased activity of c-Raf and decreased cell proliferation.
