3-Aminoquinoxalin-2-carboxylic acid is a quinoxaline derivative with potent, selective, and reversible inhibition of the amyloid precursor protein (APP) and the γ-secretase complex. It binds to the hydrophobic pocket in the γ-secretase complex, which contains residues that form an orthogonal pharmacophore model. The 3-aminoquinoxalin-2-carboxylic acid has been shown to inhibit APP and γ-secretase activity at low micromolar concentrations. The 3-aminoquinoxalin-2-carboxylic acid is selective for APP over other proteins in the brain and does not inhibit other enzymes such as acetylcholinesterase or butyrylcholinesterase.
