AZ 5104 is a potent antitumor agent that inhibits the activity of human epidermal growth factor receptor (HER) and HER2. It also prevents the activation of downstream signaling pathways, such as phosphoinositide 3-kinase, nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), and mitogen-activated protein kinases. AZ 5104 has been shown to inhibit proliferation in a number of solid tumours, including breast cancer, ovarian cancer, colorectal cancer, and non-small cell lung cancer. AZ 5104 binds to the HER receptor and blocks its activation. This inhibition leads to an increase in apoptosis through the induction of DNA fragmentation. AZ 5104 also has pharmacokinetic properties that are similar to those of osimertinib, which is used for advanced renal cell carcinoma.
