N-(2-Aminophenyl)-N'-phenylheptanediamide (NAPHA) is a non-competitive inhibitor of histone deacetylase (HDAC). It has been shown to be an effective drug for the treatment of diseases such as congestive heart failure, lymphocyte transformation, and nervous system disorders. NAPHA inhibits the activity of HDAC and prevents the removal of acetyl groups from lysine residues on histones. This inhibition leads to increased levels of acetylated histones, which in turn increases the transcription of genes that are associated with cell proliferation and differentiation. The hydroxyl group on NAPHA allows it to form a covalent bond with the enzyme, resulting in a more stable complex that does not dissociate easily. NAPHA also competes with other substrates for binding sites on HDAC enzymes and therefore decreases cellular levels of these other substrates.
