2-Amino-3-bromo-5-chloropyrazine (2ABPC) is a hydrophobic, nucleophilic, and reactive nitrile that can be used to synthesize pharmaceutically active compounds. It is synthesized by the reaction of 2-aminopyridine with bromochloromethane in the presence of sodium methoxide. The synthesis of 2ABPC is highlighted in the scheme below:
The first step in this process involves the chlorination of 2-aminopyridine to produce 2-(chloromethyl)pyridine. The second step is a regioselective nitration using bromochloromethane in the presence of sodium methoxide to produce 2-(chloromethyl)-5-nitropyridine. Finally, reduction with hydrogen gas and hydrochloric acid produces 2ABPC.
