L-Aspartic acid 4-tert-butyl ester is a peptidomimetic that inhibits the enzyme glycogen phosphorylase. This compound has been shown to be an effective inhibitor of the enzyme in assays and yields, as well as being a good substrate for the enzyme. L-Aspartic acid 4-tert-butyl ester is synthesized by reacting l-aspartic acid with benzoyloxymethyl chloride and chloroformate. The synthesis of L-aspartic acid 4-tert-butyl ester requires a chlorinated solvent such as chloroform. The reaction requires a base to deprotonate the carboxylic acid moiety, which leads to the formation of an enolate intermediate that reacts with the methyl halide to form the product.
