N-alpha-Acetyl-L-tryptophan 4-nitrophenyl ester is a fluorogenic substrate for serine proteases. It is used to study the catalytic mechanism of serine proteases and has been shown to be an excellent probe for kinetic studies. The reaction is performed in an organic solvent, such as N,N-dimethylformamide or dichloromethane. The reaction proceeds through a series of steps that are dependent on the substrate binding site, the rate of fluorescence emission and the catalytic mechanism. It follows an exponential homologous kinetics with a constant kcat and Km.
