Acetyl-O-tert-butyl-L-threonine is a synthetic, cyclodepsipeptide analog of threonine. It is synthesized by the cyclization of acetylated oxazolones. Acetylation at the C4 position creates a chiral center and this molecule can exist in two stereoisomers: S or R. The biological activity of these stereoisomers is different. The R-isomer has higher affinity for the membrane surface and better permeability through the cell membrane than does the S-isomer. This difference in permeability may be due to the orientation of hydrophobic groups on the molecule. Acetyl-O-tert-butyl-L-threonine has been shown to have pharmacokinetic profiles that are advantageous for drug delivery as well as interactions with other drugs such as warfarin and phenytoin.
