N-alpha-Z-L-asparagine methyl ester is a methyl ester of asparagine that is used to assay the protease specificity of pancreatic enzymes. N-alpha-Z-L-asparagine methyl ester is added to a trypsin or proton substrate and the hydrolysis products are analyzed by thin layer chromatography. This technique has been used in the study of various proteases, including trypsin, chymotrypsin, elastase, and carboxypeptidase A2. The plate assay can be used to determine the family of enzyme (proteolytic or amidolytic) from which a particular enzyme may derive. N-alpha-Z-L-asparagine methyl ester has also been found to have esterase activity against glycerol esters, butyrate esters, and propionate esters.
