Description:

Ac-Phe-Tyr-NH2 is an amide of the amino acid phenylalanine and tyrosine. It is a potent inhibitor of pancreatic lipase, which is responsible for the hydrolysis of triglycerides to monoglycerides and fatty acids in the intestine. Ac-Phe-Tyr-NH2 has been shown to bind to the active site of pancreatic lipase, preventing it from binding to the substrate. This effect was found to be reversible with time, which suggested that Ac-Phe-Tyr-NH2 may not be a competitive inhibitor. The kinetic data obtained from this study indicated that Ac-Phe-Tyr-NH2 binds in a constant manner and interacts with both substrate and enzyme at the same time. A deuterium isotope effect was observed, suggesting that there is hydrogen bonding between Ac-Phe-Tyr-NH2 and substrate, which leads to an increase in binding affinity.
