5-Amino-1,4-benzodioxane is a pyrazolopyrimidine that has been shown to have antimalarial activity and to inhibit the growth of tumour cells. It has also been shown to be an effective chemotherapeutic agent for cancer treatment. 5-Amino-1,4-benzodioxane binds to the active site of GTP cyclohydrolase I (GCH1), which is a key enzyme in folate metabolism. This binding prevents the synthesis of tetrahydrofolate from 5,10 methylene tetrahydrofolate and ATP, leading to inhibition of DNA synthesis and cell division. 5-Amino-1,4-benzodioxane binds to other molecules containing hydroxyl groups such as phenolic compounds and ligands, which may account for its anti-malarial activity.
