Z-Ala-Ala-Leu-pNA is a synthetic tetrapeptide that contains serine protease activity. It is used as a substrate for the determination of proteolytic enzyme activities in muscle tissue and as a substrate for protein inhibitors. The amino acid composition of Z-Ala-Ala-Leu-pNA is different from that of the natural substrate, which provides kinetic advantages in some enzyme preparations. Z-Ala-Ala-Leu-pNA has been immobilized on sephadex g100 and used to measure serine protease activity in the extracellular environment. This synthetic substrate has also been used to characterize serine proteases by sequencing their sequences.
