3-Amino-2,6-difluorobenzonitrile (3ADFB) is an organic compound. It is a magnetic resonance imaging agent that can be used to diagnose brain tumors and other diseases. 3ADFB is synthesized by nitration of 2,6-difluoroaniline with nitric acid and fuming sulfuric acid. The product is then reacted with carbonate and stannous chloride to form the final product. The resulting imine group on the molecule reacts with protons in water to produce a protonated form of 3ADFB called TMS that has a higher solubility in water than the neutral form. This allows for more efficient use of the compound as an MRI contrast agent. Magnetic resonance spectroscopy studies have shown that the protonated form of 3ADFB has better signal intensity than its neutral counterpart due to increased lipid solubility.
