1-Oxa-3,8-diazaspiro[4.5]decan-2-one is a blocker that has a potent inhibitory effect on the binding of diazepam to the benzodiazepine site of the GABA receptor. 1-Oxa-3,8-diazaspiro[4.5]decan-2-one also inhibits chloride currents and depolarizes neurons. This drug has been shown to have an inhibitory effect on cellular effects in rat cortical cells, including inhibition of Ca2+ influx and potassium efflux and inhibition of excitatory neurotransmitter release from synaptic vesicles. 1-Oxa-3,8-diazaspiro[4.5]decan-2-one also enhances antiamnesic activity in rats by inhibiting acetylcholine release from cholinergic nerve endings.
