2,2-Bis(hydroxymethyl)-1-azabicyclo[2,2,2]octan-3-one (BHMA) is a small molecule that was originally identified as a mutagen. It has been shown to induce apoptosis in human cancer cells and to inhibit the growth of xenograft tumors in mice. BHMA binds to DNA and inhibits the expression of genes involved in the cell cycle. This leads to an accumulation of cells in the G1 phase, which triggers apoptosis. BHMA also induces autophagy, which causes cells to digest their own components and catabolize intracellular organelles such as mitochondria. Autophagy is a pro-survival mechanism that may be activated when cancer cells are under stress or otherwise threatened with death. Caspase-independent cell death induced by BHMA is characterized by mitochondrial membrane potential loss and release of cytochrome c into the cytosol
