2-(4-Bromophenyl)acetohydrazide (BPAA) is a competitive inhibitor of the enzyme phosphatase. BPAA has been shown to inhibit the phosphatase activity in human intestinal cells and in other animal models. The molecular modeling studies of BPAA have revealed that this compound binds to the active site of the enzyme, which is a phosphoryl group with a hydroxyl group on carbon 2. The orientation of these groups are such that they are oriented towards one another and hydrogen bonds can form between them. This orientation rationalizes the inhibition by BPAA, which dephosphorylates L-phenylalanine, an amino acid necessary for protein synthesis. Inhibition of phosphatases by BPAA may be used as a therapeutic strategy against diseases associated with excess production of inflammatory cytokines such as nitric oxide synthase.
